В Государственную фармакопею Российской Федерации XV изд. (ГФ РФ) включена фармакопейная статья (ФС) на фармацевтическую субстанцию бисопролола фумарата, однако ФС на лекарственный препарат отсутствует. Зарубежные фармакопеи устанавливают разные требования к контролю качества лекарственных средств бисопролола фумарата. В связи с этим представляется целесообразным систематизировать информацию по национальным и зарубежным фармакопейным требованиям к лекарственным препаратам и фармацевтическим субстанциям бисопролола фумарата.</p></sec><sec><title>ЦЕЛЬ</title><p>ЦЕЛЬ. Систематизация требований к качеству лекарственных средств для подготовки рекомендаций по составлению спецификаций на препарат бисопролола фумарата в лекарственной форме «таблетки» и фармацевтическую субстанцию бисопролола фумарата.</p></sec><sec><title>МАТЕРИАЛЫ И МЕТОДЫ</title><p>МАТЕРИАЛЫ И МЕТОДЫ. В работе использовали методы сравнительного информационно-аналитического исследования и контент-анализа. Проведено исследование требований ГФ РФ XV изд. и монографий следующих фармакопей: Европейской (Ph. Eur.), Британской (ВР), США (USP), Китайской Народной Республики (ChP), Японской (JP), Республики Корея (KP) и Индийской (IP) к качеству лекарственных средств бисопролола фумарата и материалов регистрационных досье.</p></sec><sec><title>РЕЗУЛЬТАТЫ</title><p>РЕЗУЛЬТАТЫ. ГФ РФ и зарубежные фармакопеи Ph. Eur., USP, IP, ChP, JP и KP содержат ФС и монографии на фармацевтическую субстанцию бисопролола фумарата. На препарат бисопролола в лекарственной форме «таблетки» отсутствуют национальные стандарты качества, монографии на данную лекарственную форму включены в BP, USP, ChP и JP. Проведен сравнительный анализ ФС ГФ РФ и монографий зарубежных фармакопей. ФС «Бисопролола фумарат» ГФ РФ XV изд. в целом гармонизирована с требованиями Ph. Eur., однако имеются различия в части подтверждения подлинности действующего вещества и необходимости контроля фумаровой кислоты. Изучены материалы регистрационных досье на фармацевтические субстанции и препараты бисопролола фумарата. Обоснована необходимость «гибкого подхода» к контролю органических примесей в фармацевтических субстанциях и лекарственных препаратах, проведению испытания на растворение, возможность использования альтернативных комбинаций методов высокоэффективной жидкостной хроматографии (ВЭЖХ) и тонкослойной хроматографии либо ВЭЖХ и спектрофотометрии для препарата.</p></sec><sec><title>ВЫВОДЫ</title><p>ВЫВОДЫ. В результате проведенного исследования обоснован выбор показателей качества и методов анализа для идентификации, испытания на растворение для подготовки рекомендаций по составлению спецификаций на препараты бисопролола фумарата в лекарственной форме «таблетки». Установлены примеси, относящиеся к технологическим примесям синтеза фармацевтической субстанции и продуктам деградации, что также может быть использовано для подготовки рекомендаций по составлению спецификаций. Обоснована необходимость оценки и контроля возможности образования генотоксичных примесей при получении фармацевтических субстанций.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>INTRODUCTION</title><p>INTRODUCTION. Beta-blockers are one of the major groups of medicines used to treat cardiovascular diseases. Bisoprolol fumarate is of particular interest as it shows high selectivity towards beta-1 receptors. A monograph on bisoprolol fumarate drug substance was included in the State Pharmacopoeia of the Russian Federation, Edition XV (SP RF); however, there is no monograph on the medicinal product. Thus, it seems advisable to systematize the data on Russian and other national compendial requirements for bisoprolol fumarate medicinal product and drug substance.</p></sec><sec><title>AIM</title><p>AIM. This study aimed to systematize quality requirements for drug substances in order to develop the guidelines on specification drafting for bisoprolol fumarate drug substance and bisoprolol fumarate tablets.</p></sec><sec><title>MATERIALS AND METHODS</title><p>MATERIALS AND METHODS. The study used a comparative analysis and a content analysis. The study object included the monographs of SP RF, European Pharmacopoeia (Ph. Eur.), British Pharmacopoeia (ВР), United State Pharmacopeia (USP), Pharmacopoeia of the Peoples’ Republic of China (ChP), Japanese Pharmacopoeia (JP), Korean Pharmacopoeia (KP), and Indian Pharmacopoeia (IP) for the quality of bisoprolol fumarate drug substance.</p></sec><sec><title>RESULTS</title><p>RESULTS. Monographs for bisoprolol fumarate drug substance were included in SP RF, Ph. Eur., USP, IP, ChP, JP, and KP. There was no quality specification for bisoprolol fumarate tablets in SP RF; however, the monographs for this dosage form were included in BP, USP, ChP, and JP. The monograph Bisoprolol fumarate (SP RF) was compared to other pharmacopoeias; it is generally harmonized with Ph. Eur.; however, the monographs differed with regard to authentication of the active substance and the need to control fumaric acid content. Registration dossiers were analyzed for bisoprolol fumarate drug substances and tablets. It was concluded that combinations of high-performance liquid chromatography-thin-layer chromatography or high-performance liquid chromatography-spectrophotometry can be used to assess the quality of bisoprolol fumarate and identify the active substance.</p></sec><sec><title>CONCLUSIONS</title><p>CONCLUSIONS. The performed study has justified the choice of quality parameters and analytical methods used to develop the guidelines on preparing specifications for bisoprolol fumarate tablets.
