В основном в научной литературе рассматривается проблема выбора дозы лекарственного средства для инициации клинических исследований (1 фаза). При проведении доклинических исследований также необходимо решать вопросы о выборе доз для изучения токсических и фармакологических эффектов препаратов. Основные подходы, которые применяются для трансляции доз в клинические исследования, могут быть применимы также для выбора и обоснования доз при планировании и проведении доклинических исследований. Представлен обзор основных методов, которые могут быть использованы для выбора и обоснования доз при планировании и проведении доклинических исследований: межвидовой перенос доз с применением коэффициента, учитывающего разницу в площади поверхности тела; рассмотрены случаи, когда возможен прямой перенос доз в пересчете на единицу массы тела; отдельное внимание уделено вопросу межвидового переноса доз на основании данных фармакокинетических исследований. Отмечено, что универсального подхода для межвидовой трансляции доз не существует, всегда необходимо учитывать весь объем информации относительно изучаемого препарата, в том числе особенности его химической структуры, способ его планируемого применения, фармакокинетические параметры, данные доклинического и клинического изучения фармакодинамических параметров, наличие межвидовых особенностей фармакокинетики и фармакодинамики.</p></abstract><trans-abstract xml:lang="en"><p>One of the major obstacles to effective translational medicine is the challenge of translating animal research results into clinical studies. Scientific literature mainly addresses the selection of the drug dose at initiation of clinical trials (Phase 1). Appropriate selection of doses is also essential for preclinical toxicology and pharmacology studies. Some basic principles that are used when translating dosages from animal models to humans are applicable to selection and justification of doses when planning and conducting preclinical studies. The paper provides an overview of the main methods that can be used for selection and justification of animal doses in preclinical studies, e.g. cross-species dose conversion using body surface area scaling. It summarises situations when doses may be directly converted based on body weight. The paper gives special attention to cross-species dose translation according to pharmacokinetic data.
